BACKGROUND: Early knee osteoarthritis (KOA) is difficult to diagnose due to the limited sensitivity of conventional imaging and the heterogeneous presentation. This exploratory study aimed to characterize patients with early KOA symptoms by integrating arthroscopy, MRI, and synovial histopathology, and to assess the complementary value of these modalities. METHODS: We included 130 adults (> 18 years) with knee pain lasting > 3 months and inconclusive clinical and radiographic findings. Exclusion...

Dendrimeric Antigens for Passive Mast Cell Activation in the Evaluation of Amoxicillin Allergy

Autores:

Amene Tesfaye Ayane, Maria Salas, Sara Benede, Maria J Rodriguez-Sanchez, Maria J Rodriguez-Sojo, Isabel M Jiménez-Sánchez, Gador Bogas, Ezequiel Perez-Inestrosa, Cristobalina Mayorga, Juan L Paris, Alba Rodríguez-Nogales, María J Torres, María I Montañez

CONCLUSIONS: DeAns are effective platforms for investigating effector cell activation in AX allergy. By fine-tuning structural attributes-size and multivalence-we reveal the promising utility of DeAns in pMAT that leverage commercial cell lines and patient sera. This approach could address key limitations of β-lactam allergy diagnostics, enabling more reliable and standardized in vitro testing.

Clinical and serological associations of interferon-α and interferon-γ in rheumatoid arthritis patients

Autores:

Fuensanta Gómez-Bernal, Juan C Quevedo Abeledo, Cristina Almeida-Santiago, Yolanda Fernández-Cladera, Enrique García-Barrera, Luisa M Villar, Raquel Largo, J Gonzalo Ocejo-Vinyals, Miguel Á González-Gay, Iván Ferraz-Amaro

CONCLUSION: Serum levels of IFN-α, but not IFN-γ, are associated with disease activity in patients with RA. However, neither IFN is correlated with the cardiovascular comorbidities commonly observed in this population.

Dual Action of Dipyridamole in Experimental Rheumatoid Arthritis: Suppression of Joint Inflammation and Upregulation of Muscle Anabolism via Adenosine and AMPK Pathways

Autores:

Miguel Marco-Bonilla, Maria Fresnadillo, Irene Sanchez-Platero, Macarena de la Riva-Bueno, Fernando Huete-Toral, Gonzalo Carracedo, Carmen Conde, Yolanda Benitez, Pablo Minguez, Sandra Carolina Cifuentes, Joaquin Rams, Gabriel Herrero-Beaumont, Raquel Largo, Aránzazu Mediero

CONCLUSION: These results support a dual role for dipyridamole in RA, combining robust anti-inflammatory effects with a novel, myostatin-linked anabolic action on sarcopenia, mediated through adenosine and AMPK signaling, distinct from conventional therapeutic mechanisms.

High-Dimensional Immune Profiling of Human Retinal Detachment Samples Using Spectral Flow Cytometry: A Protocol for Intraocular Immunotyping

Autores:

Retinal detachment (RD) disrupts the eye's immune-privileged status, causing a local inflammatory response that contributes to adverse clinical outcomes, including proliferative vitreoretinopathy and suboptimal visual recovery. Comprehensive profiling of intraocular immune cells will offer mechanistic insights and support the development of personalized immunomodulatory strategies. Here, we describe a robust and standardized protocol for the collection and high-dimensional analysis of the...

Distinct Effects of Complement C4A and C4B Copy Numbers in Systemic Sclerosis Serological and Clinical Subtypes

CONCLUSION: This study highlights distinct genetic contributions of C4A and C4B in SSc subtypes susceptibility. Our findings suggest that lower C4 CNs, particularly C4A, increase the risk of the severe dcSSc subtype, potentially through a mechanism involving immune complex clearance.

Increased expression of Toll-like receptors and associated alarmins in temporal arteries of patients with giant cell arteritis

CONCLUSION: TLRs are overexpressed at the site of vascular inflammation in C-GCA and at a lower level in EC-GCA and PMR with negative TAB. Moreover, HMGB-1, SAA, and fibrinogen may serve as disease biomarkers of patients with C-GCA.

Galectin-1 and -9 (Gal1/9) are essential mediators of immune-inflammatory responses, which makes these proteins potential biomarkers for immune-mediated diseases (IMIDs), such as rheumatoid arthritis (RA), psoriasis (PS), psoriatic arthritis (PsA), inflammatory bowel disease, and systemic lupus erythematosus (SLE). Our aim was to evaluate plasma Gal1/9 differences between IMID patients and healthy donors (HD). We analyzed 980 plasma samples divided into two analytical cohorts (600 discovery...
